3. Solution preparation
3.1 Standard preparation:
Weigh accurately about 10mg of PC and 10mg of PS in 25ml volumetric flask, dissolve and make up the volume with n-hexane: isopropyl alcohol(80:20,V/V),constant volume,get the Standard I, Draw accurately about 5ml Standard I in 10ml volumetric flask,constant volume, get the Standard II.
3.2 Sample preparation:
Weigh accurately about 10mg of test sample in 25ml volumetric flask,dissolve and make up the volume with n-hexane: isopropyl alcohol(80:20,V/V),diluting to volume with n-hexane: isopropyl alcohol.
4. Procedure
In the above chromatography conditions, After the baseline smoothly, inject the Standard I 3 times, peak area relative standard deviation (RSD)<3.0%.Then in the following order into the sample determination:Standard I- sample solution-Standard II- sample solution -Standard I, The retention time of PC is about 10 min, The retention time of PS is about 14 min.
5. Calculation
Using external standard two point method logarithmic equation calculation content.
With the reference substance solution 1 and 2 reference substance solution peak area, the abscissa denotes the logarithm,Reference substance concentration logarithm to establish standards for the vertical curve equation:lgY=a lgX+b,R>0.999,a is the Coefficient,b is the intercept.Sample of the peak area into the equation of the concentration of PS Y of the sample drawn.
