2.4 1% trifluoroacetic acid solution: take 1ml trifluoroacetic acid solution, dilute with grade 1 water to 100ml, go with 0.45μm filter membrane.
2.5 HPLC instrument with UV detector.
3. Chromatographic condition
3.1 The chromatographic column:
Alltech prosphere C4 300A(150 mmx4.6 mm),
Alltech All-guardTM Guard(7.5mm x4.6mm), or same function column.
3.2 Detect wavelength: 280nm.
3.3 Flow rate: 1.0ml/min.
3.4 Column Temperature: 35°C.
3.5 Injection Volume: 50μl.

4. Test steps
Take sample and lactoferrin standard reference 0.1g each, both add 10ml grade 1 water (or NaCl solution), and go through 0.45μm filter membrane, then we get the sample solution and the reference solution. Use 50μl each to test at 280nm, and make the same remaining time (about 10min). The main peak is the peak of Lactoferrin. Calculate all the peak area among the twice remaining time. The ratio of main peak area and all peak area is the lactoferrin assay out of the total protein.
